Been looking and I think peptone is a general term for any of the variuos water soluble compounds formed in digestion and tryptone is a more specific term relating directly to componds formed by pancreatic digestion with the aid of trypsin. I'll follow the lead of my peers and I'll make a broth with chicken and a broth with the tryptone and compare results.

Has anyone tried chicken necks? Usually I boil up the chicken wing dissection leftovers from year 8 for this prac as they have been on around the same time. Unfortunately the teachers are now working on the new curriculum so everything has changed!! No chicken wing pracs in conjunction with Pasteur's...

Can't imagine it wouldn't work. Still chicken, you still make a stock from it. Really no difference in the end product.

An old topic but we have come to the same conclusion that neither the stock cubes or store bought stock works after many years of failures .

This year ,HT has suggested making broth from chicken necks .
We don't have Pepsin so Ian's recipe won't happen this round

Do you just boil necks in water or do you have to add other stuff to it as well ?
( I'm used to seeing my wife make chicken stock at home and its got vinegar , onions and a whole bunch of herbs and stuff )

It's been a while since I last did this, but I don't remember adding anything to it (See the RosalieM comment further up - that's me ). I just threw the chicken wings in and boiled it for ages.

I couldn't remember my password after 8 years out of the game and obviously don't have access to my old school email and nobody would reply to my admin requests on here so I created a new account for myself when I started working again this year!

I had to do the same thing a few years ago. Left labbie land in 2012 but returned in 2014. Couldn't remember password, hadn't planned on coming back and didn't have access to my old email. Easiest thing to do was create a new account. Hence, why I'm now "Merilyn1".

And Bigmack - think you only need the other stuff if you're planning on eating it

whats going wrong ?

this is the 6th year this hasn't worked .
This time I did chicken broth ...but still nothing grows ....or does it ??

What exactly is ment to grow and be seen in the flasks open to the atmoshere ?

All six were looking exactly the same .
Two were closed sealed tubes , two open , and two with swan necks and water .

we placed them in the prep room , by open windows , even oin the incubator after some really scungy Agar plates were removed .

So heres a Pic of what we have . Hoping someone can see something obviously wrong that we are doing .

That's really strange. We use a tried and true recipe I inherited from the previous labbie here and it never fails. It is almost identical to the recipe on the ASSIST website for agar plates (minus the agar of course!!!)
We have a set up that has been going strong since 2012.

here are ours.
I just add a teaspoon of vegemite in water and shake.
It has never let me down.
The students boil the water and as you can see it works a treat.
we placed it in front of the window.

yes we also left them for a week.
i have used vegemite only for nearly 10yrs and never had a fail.
we used 500ml conical flasks but in the past we have also used 250ml.
someone prior to me made the "S"bent tubes. they are 7cm wide and longest is 11cm long (goes into plug) and other end is 6cm high.

did you buy the "S"tubes or did someone make them?
maybe try making some and yes they are the largest westlab stoppers

I made the "S' tubes .
I would understand that nothing would grow in a flask with an "S" tube .( with water in the bend , its airtight)

The issue we have is that nothing seems to grow in the straight tubes which are open to the air borne nastys.

LOL, I was starting to think that this worked well for Lewy as back in the day ,there was bugger all sanitation and horses would crap in the streets ....the air would be full of nasty things whereas nowadays , we are so hygiene crazy that the air contains little .

You might have to walk past them and cough on them a few times a day

I'll 'fess up. A few years ago, at a different school, the Bio teacher and I were perplexed why the swan neck tubes had growth and the straight tubes didn't. We may have done a bit of a swap around
