No mention of calibration at all in the instruction manual. The closest is it says to use a "melt
indicator inside a small glass vial" and that indicator tape is insufficient.

We also have the 'all american' autoclave, but I am also unsure if it needs testing. We have a compliance officer at school that said it needs calibration. I phoned/emailed everyone that has to do with autoclaves I could. Most never replied and the one that did say it didn't need to be done.

Haven't actually used it, but you could try water balloons. Might be a bit harder to keep from bursting but it wouldn't have to break your budget with the cost of eggs these days.

I'm not totally sure, but I would use it like this (not to scale :D ). This is to remove the sample from direct heat from the bunsen at the bottom of the flask. You can see on your picture that it has a release vent to prevent the flask from becoming too pressurised.

I just phone around the local butchers. I find at least one willing to help. They just generally need more time to get you order together so you need 2-3 week notice for dissection pracs.

Thanks. Never thought about a dye alternative. Been getting the food dye from the supermarkets as that is what they had when I got here. Now it wont be so annoying when the students (and teachers) use half a bottle of dye to dye something that just takes a few drops.

I always make it in a schott bottle in the autoclave.
Steps/warnings:
Quick initial mix
Make sure you a bottle at least double your volume (eg. 1L bottle for 500ml of agar)
Tighten the schott bottle to just tight, then turn the lid back a quarter turn.
Indicator tape
Autoclave
Cool and pour.

The simple answer is you are adding the Nitric acid to decrease the pH of the solution as well as increase the concentration of No3 to prevent the precipitation of hydrolysed iron. If you want to make it fresh, you would have to make it just before they used it as even an hour before is not fresh ...

Yes, a water bath would suffice. I use a crystallising dishes for my water bath. Alternatively you can use sand instead of water, but would be far too messy with the students doing it.

10mls of broth is plenty, you just need 50 μl for each plate to spread. You will need the broths again for the second part, the student experiment. I do find the broths from Southern Bio hit and miss though.

We also had poor growth on our plates for a few years, tried a number of different things to fix it. Ended up changing the supplier of our broths and instantly perfect plates.
