Hi, just logged on to post & saw this line. I am having problems with agar setting for Yr 11 model cell prac. Have done this expt for a number of years with no problems until this year. Have repeated 3 times with same agar 1000gel varying NaOH & pp with same crumbly result. Am about to try with a different agar that is apparently around 500-600 gel strength from a different supplier & was already in my store, so will let you know how it goes. Any feedback on this would be much appreciated I am bascicaly using Robbie's method from 2006, except that I added conc pp in ethanol the first time & pp powder the 2 & 3 times. Not sure what brand agar I had in previous years as I don't have consistent supplies. Thanks

Please do ring your supplier, as soon as you can, you may find that batch is no good. The company returned all four of my agar gel 1000 as it just would not set. This new batch sets just fine.

Can I ask what is probably a dumb question.......is peptone bacteriological the same as nutrient agar?? or even anything like it!!

Peptone is an ingredient when I make my nutrient agar, along with some bonox and salt.

10g Bonox
10g peptone
5g sodium chloride
1 litre distilled water
Boil it all up to make a broth, filter, then adjust the pH to 7.2 (I forgot to do that last time and it was all good).
Add 15g of agar, make the plates as per usual.

re peptones; they are derivatives of proteins which provide extra amino acids for bugs to grow within the agar structure if that makes sense.

Secondly I have finally got my agar blocks to gel correctly!! It was a specific label agar as a different one worked (which in fact only had a gel strength of 500-600, go figure!) I haven't complained to the supplier as I purchased them over 2 years ago

Thanks Anne
