Nutrient Broth

Hi All,

I have a question for you,

Does anyone know how to perpare a Nutrient Broth?

If so could you tell me what is needed to prepare this.

Obviously Biology is not my strength..

Hi Robb,
I have 2 recipes for nutrient broth;

boil 500mL distilled water, add 1/2 beef stock cube, 1/4 teaspoon vegemite and 1/2 teaspoon of peptone

The other recipe if for Tryptone soya broth
0.65g tryptone soya broth base/ 50mL distilled water
in a 100mL conical add broth base
add 50mL distilled water
boil media until base disolves
allow to cool slightly & pour broth into 4 x 10mL capped tubes
label, then sterilise at 121oC for 15mins
store in fride for use

Neutrient Broth

Dear Robb,
I have only been here a short time, but was beginning to think that there was nothing that you did not know! (and am appreciating your helpfulness)

My Nutrient Broth is similar to Liz's, but with Salt and Bonox instead of Beef Stock cube.

Being from a Physics background myself, I get a kisk out of measuring everything to a million decimal points of accuracy, rather than using teaspoons, although in truth, the accuracy is pretty irrelevant.

Ingredients
5g Peptone
5g NaCl
2g Vegemite
1g Bonox
1L Distilled Water

Method
Add solids to water and boil until all solids are disolved, (I use the hot plate with the magnetic stirrer)

If I am needing to make Nutrient Agar, I then dissolve 15g of Agar powder in the broth and pour into conical flasks, stoppered with cotton wool, and autoclave at 15 psi for 15 minutes.

This method seems to work for me, except for growing Yoghurt. That needs a Lactose based solution, I think. (eg McCockney's Agar

Hi Colleagues,

Thanks for your introduction in your topic post Ian, It has been an interesting moment for me, I have my Chemistry and Physics Double Degree and Masters in Chem but, as you can see Biology eludes me.

It is good to be asking the questions and to have the help that is always there from everyone, is certainly a credit to each and every one of you.

As you can see eventually we all get stuck at one point or another..

Hi Rob,

We actually purchase Nutrient Broth Powder from Science Supply. I've been using it for the last 3 years & works well. My days are very busy so any shorts cuts I can take I will.
