Emzyme Test

In the past I have pureed liver and placed in various buffers then put different strength Hydrogen Peroxide on it so they could graph results in a prac exam.
Terribly MESSEY
Does anybody have a better / simpler or other material than liver to do test on?

Dear Graeme (Ocker),

Hi there,

we use potato cores of same diameter and length, placed into testtubes of solutions of peroxide at different pH (6% peroxide is naturally pH3 and by adding sodium hydroxide dropwise (first use a 0.1M solution, then to get the pH 9 and 11 you will need about 2M, otherwise the volume becomes too great), the pH can be increased to Ph5, pH7, pH9 and ph11: then using hydrochloric acid the pH can be lowered to Ph1). Adding a few drops of detergent helps to 'stabilise' the oxygen foam formed, which can then be measured with a ruler.

Hi,
Our Senior Biology class did an enzyme prac the other day and used ground spinach and H2O2.
Apparently it worked really well.

Hope this is of some use.

We also use potatoes .. this one part works well.. the bubbles are well formed andf the results are repeatable...

BUT I have a question:
One teacher used Universal Indicator to adjust the Ph of the solutions..
when I checked with the pH probe.. the pH results given by the Universal Indicator were all false....
What is the reaction here? Does the peroxide react with the indicators in the Universal ?????
The chem teacher here doesnt know - and they are all looking to me to give them a reason - and...well guys :-o

We had an enzym prac test resently, we used capsicum extract (blended in mixer) and H2O2, yellow and green, green has the enzyme whereas the yellow one doesnt, its really cool and worked fabolously....

I can email you the prac if you like, and some of our results too!

Am having massive issues with my computer, it has now been taken away for repairs, so at the moment I cannot access anything :mad: As soon as I get my computer back, which was suppose to be at 12.30 yesterday, I will email/print&fax the enzyme prac for you!

After your suggestion last year I trialed slicing potato and taking slugs with a cork borerIt worked wonderfully and we used it in prac exam.
As I had over 200 students doing prac exam you can imagine how busy I was and forgot to Thank You!

Afternoon everyone!

My computer is back and well, woohoo! But as it is so hot here, I feel very lazy, so instead of emailing etc I will just post it here :thumbup:

It is activity 2 you are interested in! Please ignore my scribbles on page six, instead look for the results in the excel file. We actually wanted the graph to plataeu, so it should have increased to a certain point when it evens out, it is too hot for me to think about the actual explanation for this today, something with the active enzyme and optimal the substrate concentration, although we did not get these results this year, so for next year we are playing around with different sze test tubes and also perhaps different mL of H2O2. By the way, we used 10% H2O2.
