I mentioned a while ago using gelatine. However, I use it as a substitute for peptone not as a substitute for agar. (We have plenty of gelatine and peptone is expensive.) Both are used as a source of protein.

I don't think gelatine would be a good substitute for agar because gelatine needs refrigeration to set, whereas agar sets (and stays set) at room temperature provided you use enough.

The recipe I use for a class set of 30 plates is:

750 mls distilled water
15g agar
0.75g sucrose
3.7g sodium chloride
3.7g gelatine
2.5g Vegemite.

This has always worked well for us.

There must be more recipes for nutrient agar than anything else I have heard of and all apparently work

Thanks, Robbie - it was me who was asking about the possibility of substituting gelatine for agar. My (then) HT asked about it last year.

I ran out of agar last year and tried to substitute gelatine, what a disaster, it doesn't work!

To dispose of my agar plates, I put them inside an Oven bag and seal it, then put THIS inside a second one and seal it .. then cook it on 'High' for 15 mins in my microwave!
Seems to do the trick

Yes it just the best, and some one said, its good to watch then melt down. Nothing could grow after that, for sure.

Hi Guys,
I am micro trained. The dept of education state no growing of unknown Microbes. you can do everything you need to do for the sylabous using a peppercorn solution wich growes a safe bacteria to you, students and teachers. the plate can be desposed of in bleach or in home brand disinfectent then you can place in normal garbage.

Hi Kathy, can you please tell me how to make a peppercorn solution and how to use it. It sounds like a better alternative to agar plates.
